3228c Customer want to print on glossy labels.

It seems to work fine. I test printed them as "Thick". I'm wondering two things.

    1. Can I print these from a paper tray as "preprinted" or something else to make the fuser hotter.

    2. And more importantly will there be any long term negative effects on the machine, the fuser in particular?

The bypass is the only source to put thick stock. The 'preprinted' and other labels that you can set for the trays is only for reference for the users as to what type of stock should be in them. They have no effect on any settings (temp/speed).
Using a thick stock in the cassettes would most likely cause poor fusing and buildup in the fuser on the rollers. You can try a few and see if you can scrap off any toner on the outputted prints. You could also try raising the fusing temp for the trays about 5 degrees, but I wouldn't go any higher. My opinion is that if you start overcompensating for one thing you're going to mess things up for the normal stuff.

No problem, also remember to tell them that if it's a print job that they have to select the "thick" setting in the driver. "thick" setting on the copier screen (# key) has no effect on print jobs, only copy.
