Software you are using might not be compatible with TWAIN scanning. Try installing Irfanview. The other possibility is that your firewall is blocking the software from connecting to the network device. With many firewall softwares, the user does not always read the warning message that pops up the first time a software tries to access a network device. They just click block. After that, no more messages. You have to go into the firewall settings to reopen the port to the software. I have had a number of customers with McAfee firewall block TCP/IP printing.

Try using the network setup tool that loads with the twain driver. There might be more than 1 machine on the network so the driver doesn't know which machine to pick.

Is there a reason for choosing TWAIN over a more reliable method like SMB or scan to email? Outside of USB scanning, TWAIN is really all but dead since firewalls and other network security tools have become more sophisticated.

I have some customers, medical offices and attorneys, who use software that utilize TWAIN drivers from within the software in order to import documents directly to client/patient accounts. Much faster than scanning to folder or email, then locating the file and importing to the software.

This is the first logical explanation I've heard for using TWAIN in a long time...
